This is a “defeat all enemies”-style challenge. However, there's a twist: around halfway through the challenge, poison gas will fill the arena, constantly draining your health. You can heal to recover, but if you’re on a good pace and have upgraded your health, then you should concentrate on using your Focus for finishers to complete the challenge faster.
Located in Harlem, players are tasked with defeating 20 enemies in under a minute.

In the Upper East Side, players must defeat 20 enemies within 2 minutes and 15 seconds while some enemies are shielded.
- Mysterio Priority: Shielded enemies can't be damaged. Locating and defeating Mysterio quickly will disable these shields, making the enemies vulnerable.
- Mobility: Constant movement prevents being cornered by shielded enemies, allowing for hit-and-run tactics.
- Web Grabber: The gadget can be used to break enemy formations, making it easier to target Mysterio among the crowd.

4. Midtown's "The Invisible Enemy" Mysterium
In this challenge, you need to beat enemies quickly. There are two twists: firstly, you need to perform finishers to make the KO count, and secondly, enemies become invisible after a short time. In Chinatown, the objective is to kick 15 enemies off rooftops within 1 minute and 30 seconds.
- Avoid Laser: Avoid laser when the clock strikes 12. This way, you can use your focus on finishing moves instead of healing.
- Jump Kicks: A well-timed jump followed by an attack can send enemies flying off rooftops. It's essential to get the angle and timing right.
- Web Grabber: Aiming this gadget off the building's side can pull enemies into a free fall, counting towards the objective.

6. Williamsburg's "Grave Decision" Mysterium
This challenge is similar to Road Rage but takes place in a graveyard. The objective remains the same: don't get hit. However, Mysterio himself will show up around halfway through to add even more chaos to the equation.

The first Williamsburg challenge requires players to defeat waves of enemies without taking a single hit.
- Constant Movement: Staying stationary makes you an easy target. Continuously moving, swinging, and jumping can make it difficult for enemies to land a hit.

This challenge has you fighting on a bridge, with a few floating islands nearby, as enemies and traffic spawn in. Your goal is to take out all the enemies while taking as few hits as possible. Each hit will add a point to your score, so the lower your score, the better. Be cautious of shooters and lanes of traffic on the bridge that light up, indicating an incoming car.

Your challenge here is to eliminate a number of Brute enemies specifically, though other enemies will appear as well. A train will occasionally spawn and drive through the arena wherever you happen to be at the time. You get a good amount of warning by seeing the green tracks before the train itself comes, which you need to use to your advantage to bait Brutes into the tracks for easy KOs.

In Downtown Queens, players are challenged to defeat six brute-type enemies within 2 minutes.
- Brute Priority: These enemies are tougher and can take more damage. Focusing on them first can make the challenge more manageable.
- Finishers: Using finishers on brutes can quickly reduce their health, speeding up their defeat.
